Fils de Donald Sutherland et de l'actrice canadienne Shirley Douglas.

Kiefer Sutherland se destine très tôt à la carrière de comédien et tient son premier grand rôle dans Throne of straw à l'Odyssey Theatre de Los Angeles. Après avoir poursuivi son apprentissage dans divers ateliers de Toronto, au Canada, il débute à l'écran aux côtés de son père dans Max Dugan returns (1983) de Herbert Ross, puis tient la vedette d'Un printemps sous la neige (1984), un drame pour lequel il est cité au Genie Award.

Le comédien s'établit alors aux Etats-Unis où il fait sa première apparition à la télévision dans l'épisode The Mission (1985), réalisé par Steven Spielberg dans le cadre de la série Histoires fantastiques. En 1986, il connaît son premier succès international avec Stand by me de Rob Reiner. Dès lors, il alterne les genres : la comédie avec Génération perdue (1987), le western avec Young guns (1988) et Young guns 2 (1990), ou encore le thriller avec La Disparue (1993). Mais ce sont ses prestations d'étudiant en médecine, aux côtés de sa compagne à la ville Julia Roberts, dans L'Expérience interdite (1990), de lieutenant placé sous les ordres de Jack Nicholson dans Des hommes d'honneur (1992) et d'Athos dans Les Trois mousquetaires (1993) qui lui permettent de se faire connaître auprès du grand public.

En 1996, Kiefer Sutherland n'hésite pas à incarner des personnages "détestables" : un violeur récidiviste dans Au-delà des lois, un psychopathe défiguré dans Freeway (id.) et un membre du Ku Klux Klan dans Le Droit de tuer ? (id.). L'année suivante, il met en scène son premier long métrage : La Dernière cavale, un polar qu'il interprète aux côtés de Vincent Gallo. Il poursuit sa carrière de réalisateur en 1999 avec Woman wanted, un drame sentimental dans lequel il s'éprend de Holly Hunter.

Parallèlement, Kiefer Sutherland s'essaie à la science-fiction en s'illustrant dans le sombre Dark City (1998) d'Alex Proyas. Le grand public le redécouvre en 2001 en agent de la Cellule Anti-Terroriste dans la série en temps réel 24 heures chrono. Ce rôle lui vaut le Golden Globe du Meilleur Acteur. En 2003, il retrouve un rôle de "méchant" en interprétant le mystérieux interlocuteur de Colin Farrell dans Phone game (Raccroche et tu es mort), un thriller de Joel Schumacher, un cinéaste auquel il est resté fidèle.
